Abstract

Abstract

Estrogen is a steroid compound that has many benefits for the bodys physiology.Hypoestrogenic women could have periodontitis in their mouth. Hormonereplacement therapy (HRT) is a therapy that used to reduce the disturbance causedby estrogen deficiency. However, HRT has many side effects. Theoretical studyshows that isoflavones in tofu liquid waste has an element of a steroid compound. The purpose of this study was to prove the effectiveness of isoflavones in the tofuliquid waste by examining the microscopic structure, estrogen receptor expressionand MDA levels in rat mandibular bone post-ovariectomy. This research methoduse a true experimental laboratory with randomized post test control group design.Twenty-four female wistar rats were divided randomly into six groups, consistedof a group of normal rat that does’nt ovariectomized (C1), 4 weeksovariectomized rats (C2), 8 weeks ovariectomized rats (C3) but no tofu liquidwaste was given, and 8 weeks ovariectomized rats that given tofu liquid waste atthe end of the 4th week of ovariectomy with three different doses (C4 = 1.2, C5 =6; C6 = 12 ml / kg). Histopathological slide used to see the changes resulted in anumber of osteoclasts, osteocytes, periodontal ligament width, alveolar boneheight, the amount of estrogen receptor expression and MDA levels. The resultsof statistical tests show that there is a difference in the rats between six differentgroups (ANOVA, p <0.05) and there is a close relationship between the dose ofthe tofu liquid waste with all parameter. The conclusion of this study is tofuliquid waste could improve the microscopic structure, estrogen receptorexpression and MDA levels in rat mandibular bone post-ovariectomy.

Abstract

The activation of orthodontic appliance will generate mechanical force. This forceis used to depress tooth and the tissues surrounding it then stimulates alveolarbone remodeling. Alveolar bone remodeling is divided into compression andtension area. This stress becomes a signal to activate heat shock responseyielding heat shock protein (HSP) synthesis, especiallyhsp25isessentialforalveolarboneremodelingprocessitself. The aim of this study was to comparehsp25 expression in compression and tension areas. This study using guinea pigsmandibular first insicivus that given a mechanical force to see hsp25 expressionin both areas. The HSP25 expressionwas measured by counting this protein afterbeing conducted by immunohystochemistry method. It is analysed statisticallywith one way Anova with level of significant p = 0,05.

Abstract

The main microbial culprit in dental caries is Streptococcus mutans (S.mutans),virulence of which can be observed by its differential protein expression betweencaries and caries-free subjects. The success of Immunoglobulin-Y (IgY) antiS.mutans as a passive immunization agent in eliminating S.mutans has beenreported. The aim of this study is to analyze the effect of IgY anti S.mutans on theprotein expression of S.mutans isolated from caries and caries-free subjects. Eachdental plaque was collected by swabbing the buccal surface of the first lowerpermanent molar of caries and caries-free subjects. The plaques were thencultured on agar medium TYS20B. After 72 hours, the colonies from each of themwere cultured in liquid medium TYS Broth for 72 hour. Each collected bacteria(whether from caries or caries-free subjects) were grouped into control andexposure group. In exposure group, S.mutans was exposed by pre-incubated (forone hour at 37°C) IgY anti S.mutans for one hour at 37°C. Protein expression ofS.mutans was analyzed with SDS PAGE after the preparation of its antigen andBradford protein assay. Our result shows that S.mutans 41.3 kilodalton proteinexpression of caries subjects, are up-regulated in comparison to the control group.Meanwhile, the S.mutans 41.3 kilodalton protein expression of caries-freesubjects, are down-regulated in comparison to the control group. This studysuggests that IgY anti S.mutans up-regulates 41.3 kilodalton protein expression ofS.mutans in the caries subjects. However IgY anti S.mutans down-regulates 4.13kilodalton protein expression of S.mutans in the caries-free subjects.

Abstract

Specificity Protein (SP) family members are tissue specific transcription factors. They regulate a wide range of cellular function including cells growth, apoptosis, differentiation and tumor formation. This family composed of more than 25member proteins, which contain a DNA-binding domain very well conservebetween all members with three tandem zinc fingers of the C2H2 type in theirCterminal region. Recently, it has reported in mammalian genome, there are nineSp genes (Sp1-Sp9). Some previous study reported Sp3 knockout mice haveenamel/dentin layer defect. Sp4 mRNA express in E13- E16 of WT mouse incisor.Sp7 mRNA express in at 15, 17 days of post coitum and P1 (postnatal day 1). Sp6has another name epiprofin, I also worked with this genes. Sp6 mRNA expressedin early stage of tooth development to the secretory stage of ameloblast. It alsoreported Sp6 weakly expressed in mesenchymal odontoblast of the incisor. Sp6deficient mice reported delay tooth development. SP family members play animportant role in tooth development.
